Interleukin-2 (IL-2) is an interleukin, a type of cytokine signaling molecule in the immune system. It is a protein that regulates the activities of white blood cells (leukocytes, often lymphocytes) that are responsible for immunity. IL-2 is part of the body's natural response to microbial infection, and in discriminating between foreign ("non-self") and "self". IL-2 mediates its effects by binding to IL-2 receptors, which are expressed by lymphocytes. Rec. E. coli interleukin-2 for T cell culture or antibody production is supplied by GENTAUR. The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to IL2 Inducible T-Cell Kinase (ITK). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to IL2 Inducible T-Cell Kinase (ITK). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain IL2 Inducible T-Cell Kinase (ITK), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of IL2 Inducible T-Cell Kinase (ITK) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
